Divergent Effects of Inotropic Stimulation

چکیده

Mechanical function remains depressed for hours and days after even brief periods of ischemia. To determine whether the depressed function of the reperfused myocardium could be improved by inotropic stimulation, we studied segmental function during ischemia and after reperfusion using mercury-in-Silastic length gauges in 15 dogs. During coronary artery occlusion, segmental function could not be improved by inotropic stimulation with dopamine. Release of occlusion after 30 minutes of ischemia resulted in only slight improvement in segmental function (systolic shortening at 20% of control). After reperfusion, segmental function could be markedly enhanced by inotropic stimulation. The response to inotropic stimulation was similar after reperfusion after 3 hours of ischemia if the myocardium remained viable (nine dogs). When the myocardium was necrotic (five dogs), there was no improvement after reperfusion, either spontaneously or in response to inotropic stimulation. If applicable to humans, these results suggest that intractable pump failure caused by extensive but reversible ischemia could be effectively treated by reperfusion and inotropic stimulation.

Mechanism of Action of the Thyroid Hormone on the Heart

SUMMARY  The foliowing cardiac effects may be attributed to thyroxin:  1-Thyroxin augments all anaerobic processes in the body includ::ng the heart, and decreases the glycogen content of the heart ( 1, 2, 5, 27). The resistance of the heart to anoxia is increased in hyperthyroidism ( 25). 2- Thyroxin influences the cardiac weight and prevents cardiac atrophy (3, 8, 10, 11, 19, 20, 21, 30...

Significance of the endothelin ETA receptor in the haemodynamic and inotropic effects of endothelin-1 in rats.

The main aim of the present study was to investigate the direct inotropic effects of stimulation of the endothelin (ET) receptor ETA under in vivo conditions. It is well known that ETA receptor stimulation causes pronounced vasoconstriction. The ET-1-induced coronary vasoconstriction may lead to myocardial ischaemia and, consequently, to cardiodepressor effects that may mask the direct positive...

c^-Adrenoceptor-Mediated Phosphoinositide Breakdown and Inotropic Response in Rat Left Ventricular Papillary Muscles

a,-Adrenoceptor stimulation of rat left ventricular papillary muscles by phenylephrine in the presence of propranolol resulted in rapid breakdown of phosphatidylinositol 4,5-bisphosphate (PI-4,5-Pj) and a triphasic inotropic response in a concentration-dependent manner. The release of inositol trisphosphate (IP,) was maximum within 30 seconds and remained high for at least 30 minutes. The IP3 f...

Novel cilostamide analogs, phosphodiesterase 3 inhibitors, produce positive inotropic but differential lusitropic and chronotropic effects on isolated rat atria

Objective(s): Recently, we showed that some new synthetic compounds structurally related to cilostamide (4-(1,2-dihydro-2-oxoquinolin-6-hydroxy)- N-cyclohexyl-N-methylbutanamide), a selective phosphodiesterase 3 (PDE3) inhibitor, produce inotropic effect comparable to that of IBMX (3-isobutyl-1-methylxanthine), a non-selective PDE inhibitor, but with differential chronotropic effect. In this in...

Direct positive inotropic effect of acetylcholine on myocardium. Evidence for multiple cholinergic receptors in the heart.

The effects of acetylcholine and nicotine on isometric tension, in the presence and absence of atropine, hexamethonium, and endogenous norepinephrine stores, were examined in isolated cat atria and papillary muscles. Acetylcholine exerted a negative inotropic effect, competitively inhibited by atropine, in atrial and papillary muscle; in higher concentrations in papillary muscles, it produced a...
